About the tutorial: The goal of this Scipertise tutorial is to provide participants with a better understanding of the methodologies used in approaching temporally precise single-cell-resolution optogenetics. Participants will gain a better understanding of these techniques and feel more comfortable applying these techniques in their own experiments.

Session 1:

  • Why single cell resolution optogenetics?
  • ‘Cross talk’: Why can’t conventional optogenetics support single cell control?
  • Screenings for a cell body targeted opsin, defining and analyzing soma targeting (neuron culture, imaging, screening candidates, screening potential hits)
  • Validations: membrane properties, photocurrent, temporal characterization (patching, action spectra, spike probability)
  • In vitro 1-photon single cell resolution optogenetics (patching, photostimulating with a DMD)

Session 2:

  • Why does single cell optogenetics in 3D brains require 2 photon (2P) microscopy?
  • What is 2P holography? (advantages of holography for single cell optogenetics)
  • Choosing experimental parameters in slice (2P light power, speed)
  • Validating single-cell, single-millisecond control in a brain slice (patching, channel blocking, imaging)
  • Can we achieve 2P single cell control without holography?
  • Demonstrating the power of single cell control 3D (analyzing neural connectivity in a brain slice)
